Ezh2 is a member of polycomb repressive complex 2 (PRC2). This protein act as a lysine methyltransferase that catalyzes the mono-, di-, and trimethylation of H3K27 and regulates gene expression by engaging in methylation activity, in addition to maintaining pluripotency during ESC differentiation (1-4).

Function and Interaction | |
Ezh2 is a SET domain-containing lysine methyltransferase capable of catalyzing the mono-, di-, and trimethylation of H3K27, and the majority of these marks are associated with transcriptional repression (1-2). Ezh2 is regarded as an important modulator of cell differentiation. However, recent studies have shown that Ezh2 also participates in the tissue regeneration process. Ezh2 is essential for tissue repair, and deletion of Ezh2 can lead to an inability to engage in pancreatic regeneration and the promotion of KRas (G12D)-induced neoplasia (5). EZH2 is highly expressed in pro-B cells and is involved in early B-cell differentiation, but its expression declines progressively in pre-B cells and immature B cells (6). Ezh2 is thought to be related to DNA methyltransferases and histone deacetylases, indicating that these three types of epigenetic silencing machinery could all contribute to the abnormal control of gene expression in cancer cells (7-9). Ezh2 has also been shown to interact with ATRX, EED, and VAV1 (10-12).

Disease Association | |
Ezh2 is associated with head and neck squamous cell cancers (HNSCC), myeloid malignancies, prostate cancer, and breast cancer (13-18). Mutations in EZH2 can also result in Weaver syndrome (19-20).
