Introduction

Full Name: Lysine (K)-specific demethylase 5C . KDM5C is a member of the highly conserved ARID protein family and was first recognized as an X-chromosome gene that escapes X-inactivation in both mice and humans. It contains a JmjN domain, an ARID domain, a JmjC domain, a C5HC2 zinc finger domain, and a PHD zinc finger domain. KDM5C expression may be tissue- and developmental stage-specific, and it is associated with transcriptional repression and chromatin remodeling, playing a role in normal brain development and function, tumorigenesis, development, cell growth, and differentiation (1-13).

Function and Interaction

KDM5C forms a complex with HDAC1, HDAC2, G9a, and REST. REST and KDM5C have been shown to co-bind the promoter regions of some REST target genes, indicating the involvement of KDM5C in chromatin dynamics and REST-related repression (2). KDM5C is also associated with Smad3, an intracellular mediator of transforming growth factor-β (TGF-β), and acting as a corepressor of Smad3. Hence, KDM5C may be involved in tumorigenesis via interfering with the tumor-suppressing activity of the TGF-b/ Smad3 signaling pathway (3).

Disease Association

Mutations in KDM5C are related to X-linked mental retardation (XLMR) and epilepsy (4-5). One study also associated a mutation in KDM5C with a patient with autism spectrum disorder (6). This novel mutation in KDM5C can result in intellectual disability, short stature, speech delay, hyperactivity, violent behavior, and a high palate (8). KDM5Cs are suggested to act as tumor enhancers in clear-cell renal cell carcinoma (ccRCC) (7).
