Introduction

Full name: Lysine (K)-specific demethylase 5B . KDM5B displays a demethylase-containing Jmjc domain, along with PHD domains and an ARID domain. It is highly expressed in breast cancer cells, while its expression is limited in normal tissues, requiring iron and α-ketoglutarate to function. Additionally, KDM5B is involved in processes such as development, differentiation, cell proliferation, transcriptional regulation, and chromatin remodeling (1-5). KDM5B can remove mono-, di- and trimethylation on histone H3K4 (1).

Function and Interaction

KDM5B regulates the expression of many genes related to development, the cell cycle, signal transduction, and metabolism (3,6-7). KDM5B overexpression can increase the transcription of the androgen receptor, which plays a vital role in prostate cancer development via its JmjC domain (1). KDM5B can cooperate with TFAP2C and Myc in suppressing p21 (cip) (CDKN1A), which acts as an inhibitor during the cell cycle (6). This factor can be recruited by Rb to the promoter of some E2f target genes, enabling it to take part in the Rb pathway (8). KDM5B is an activator, rather than a repressor in the self-renewal network (9). It also plays a role in cell-fate decisions (10). However, KDM5B also acts as a transcriptional repressor, and its interactions with brain factor-1 (BF-1) and paired box 9 (PAX9) can dramatically increase its suppression effect (11-12). KDM5B interacts with class I HDACs, class IIa HDACs and N-CoR in breast cancer cells (4). KDM5B can also interact with TIEG1 (transforming growth factor-β inducible early gene-1) to inhibit tumorigenesis by regulating TGF-β signaling (13).

Disease Association

KDM5B is upregulated in breast cancer and is involved in regulating cell proliferation by interacting with tumor suppressor genes. KDM5Bis also upregulated in prostate cancer cells (1-2,5,12).
