Full name: cAMP-response element binding protein (CREB) binding protein.
CREBBP is a large protein consisting of over 2400 amino acids,is first isolated as a nuclear protein that binds to cAMP-response element binding protein(CREB),shares regions of very high sequence similarity with protein p300 in its bromodomain, cysteine-histidine-rich regions, and histone acetyltransferase domain,has intrinsic histone acetyltransferase activity and also acts as a scaffold to stabilize additional protein interaction with the transcription complex(1-2).CREBBP plays critical roles in embryonic development, growth control, and homeostasis(1-7).

CREBBP has intrinsic histone acetyltransferase activity towards H3 and H4(1-2).CREBBP is known to interact with a variety of DNA-binding transcriptional factors including nuclear hormone receptors,CREB,c-Jun/v-Jun,c-Myb/v-Myb,Sap-1a,c-Fos and MyoD,binds specifically to phosphorylated CREB and enhances its transcriptional activity toward cAMP-responsive genes,acts as a circadian transcriptional coactivator which enhances the activity of the circadian transcriptional activators:NPAS2-ARNTL/BMAL1 and CLOCK-ARNTL/BMAL1 heterodimers(3-8).Acetylation activity of CREBBP promotes removal of chromatin-bound PCNA and its degradation during nucleotide excision repair(NER)(9).
