Full Name: E1A binding protein p300
.
Ep300 was first identified as one of the many targets of the adenovirus E1A protein. Ep300 acts as both a histone acetyltransferase and a coactivator in some signal-associated events. It possesses three cysteine-histidine-rich domains (CH1, CH2, and CH3), a KIX domain, a bromodomain, a histone acetyltransferase (HAT) domain and a C-terminal glutamine-rich domain. It is closely related to CREBBP and exhibits some redundant functions associated with the activities of hundreds of transcription factors, cell proliferation, differentiation, and development (1-6).

Ep300 has been shown to play a role in organ development in mice, including neural, vertebral, limb, heart, skin, and hair development, and its expression is thought to be developmentally modulated in a stage- and tissue-specific manner during mouse embryogenesis. Although Ep300 and CREBBP share many common functions, their expression patterns exhibit subtle differences at the cellular and subcellular levels during embryogenesis (4). Cells lacking Ep300 exhibit defects in neurulation, proliferation, and heart development (5). The functions of Ep300 and CREBBP in F9 cell differentiation also differ (6). Signal transducer and activator of transcription 2 (STAT2) and the RelA (p65) subunit of NF-kappaB compete for binding with the first zinc finger domain of p300, thus enabling interferon-alpha (IFN-alpha) to suppress NF-kappaB-dependent HIV gene expression induced by tumor necrosis factor-alpha (TNF-alpha) (7). Ep300 is believed to be recruited by ligand-activated ER, resulting in the acetylation of ACTR, thereby destabilizing the ACTR-p300-ER complex and downregulating nuclear receptor-related transcription (8). Ep300 acetylates p53 in its C-terminal DNA-binding regulatory domain and exhibits other non-histone substrates, such as TFIIE (ocmponents of the general transcription machinery) (9-10). Ep300 acetylates promoter histones through its HAT domain, thus regulating transcription, and the bromodomain of Ep300 could modulate its acetyltransferase activity. Additionally, Ep300 can interact with transcription factors, thus regulating the expression of target genes transcribed by RNA polymerases I, II, and III (11-14). Ep300 is associated with CREB and other factors through its KIX domain, which controls CREB-related specific gene transcription (15). E1A is an adenoviral oncoprotein that binds to p300/CBP and is involved in regulating transforming growth factor (TGF)-beta signaling (16).

Mutations in p300 can cause Rubinstein-Taybi syndrome, which is characterized by facial abnormalities, broad thumbs, large toes, and mental retardation (17).
