Introduction

Full name: NAD-Dependent Protein Deacetylase Sirtuin-6. SIRT6 is a member of the sirtuin family of NAD-dependent enzymes that are implicated in epigenetic gene silencing,recombination,metabolism,cell differentiation and in the regulation of aging(1).SIRT6 contains a splayed zinc-binding domain but lacks the conserved,highly flexible,NAD(+)-binding loop,is able to bind NAD(+) in the absence of an acetylated substrate(2).SIRT6 has deacetylase activity towards histone H3K9ac and H3K56ac(1-6).

Function and Interaction

SIRT6 is a NAD-dependent protein deacetylase which has deacetylase activity towards histone H3K9ac and H3K56ac,modulates telomeric chromatin through acetylation activity of histone H3 during the S-phase of the cell cycle,deacetylates histone H3K9ac at NF-kappa-B target promoters and may down-regulate the expression of a subset of NF-kappa-B target genes(1-6).SIRT6 acts as a corepressor of the transcription factor HIF1A to control the expression of multiple glycolytic genes to regulate glucose homeostasis(7).
