Introduction

Full name:Lysine (K)-specific demethylase 4B. KDM4B is a demethylase that specifically demethylates tri-methylation on histone H3K9(H3K9me3) and is reported to activate gene transcription through regulation of chromatin structure,plays a special roles in colorectal cancer(CRC),prostate cancer,gastric cancer and pancreatic cancer,is required for expression of the ER and FOXA1 genes,involving in process like development,transcriptional regulation,cell apoptosis,cell proliferation and suppression of senescence(1,2,3,4,5,7,8,10).

Function and Interaction

KDM4B/JMJD2B was found as a direct target of hypoxia-inducible factor 1α (HIF1α) and regulated by HIF1α and estrogen receptor α (ERα) in breast cancer and promoted hormonally responsive breast carcinogenesis(1,2,3,5).KDM4B is overexpressed in colorectal cancer and is significant for the mediation of mitochondrial apoptosis in colorectal cells(2).KDM4B interacts with the transcription factor GATA-3 in BCa cell lines and directly co-activates GATA-3 activity(4).KDM4B could regulate multiple pathways which contribute to carcinogenesis,including the cell-cycle pathway(6).KDM4B silencing promotes cell apoptosis,plays an essential role in mitochondrial apoptosis by targeting HAX1(2).KDM4B is a positive regulator of EMT process promoting epithelial-mesenchymal transition through epigenetic regulation of ZEB1 in pancreatic cancer(7).KDM4B/JMJD2B is found to be necessary for self-renewal of embryonic stem cells and induced pluripotent stem cell generation(9).

Disease Association

KDM4B is shown to be overexpressed in clinical colorectal cancers and regulates cell apoptosis,it also promote growth in bladder cancer,lung cancer and gastric cancer(1,2,7),and KDM4B promotes epithelial-mesenchymal transition through up-regulation of ZEB1 in pancreatic cancer(7).
