Introduction

Full name: Lysine (K)-specific demethylase 4C. KDM4C is a member of the Jumonji domain 2 (JMJD2) family,is amplified and overexpressed in esophageal squamous carcinomas,colon cancer,prostate cancer,basal-like breast cancer(3,4,5,6,8,11),demethylates H3K9me3/me2 and H3K36me3/me2 through a hydroxylation reaction requiring iron and α-ketoglutarate as cofactors(1,9,10),involing in process like development,differentiation,cell growth,transcriptional regulation,chromosome segregation and carcinogenesis(2,5,6,7,8).

Function and Interaction

KDM4C induced demethylation at histone H3K9 to activate VE-cadherin expression and thus enabled the differentiation of mESCs to endothelial cells(2).KDM4C forms complexes with β-catenin,an oncoprotein whose overexpression is crucial for the development of most colonic tumors,KDM4C is required for efficient expression of the growth stimulatory proteins FRA1 and cyclin D1 as well as the survival factor BCL2(4).KDM4C and LSD1 interact and both demethylases cooperatively stimulate androgen receptor-dependent gene transcription,KDM4C,androgen receptor and LSD1 assemble on chromatin to remove methyl groups from mono, di and trimethylated H3K9(5).KDM4C demethylase activity regulates the expression of genes critical for stem cell self-renewal,including NOTCH1,is a driving oncogene in the 9p23-24 amplicon in human breast cancer[6].KDM4C associates with mitotic chromatin through Tudor domains and dysregulation of KDM4C demethylase activity promotes chromosome instability(CIN) by impairing the fidelity of mitotic chromosome segregation(8).KDM4C is targeted to H3K4me3-positive transcription start sites,where it can contribute to transcriptional regulation(9).

KDM4C is a crucial oncogene amplififed and overexpressed in esophageal squamous carcinomas,colon cancer,prostate cancer and basal-like breast cancer(3,4,5,6,8).
