Introduction

Full name: Chromodomain helicase DNA-binding protein 2 . CHD2 is a chromatin remodeler belonging to the CHD1-CHD2 subfamily of the SNF2 family. It shares homology with CHD1, containing two chromodomains in the N-terminal domain, a central helicase-like ATPase motor, and a C-terminal DNA-binding domain that preferentially binds to A/T-rich sequences involved in regulating development, hematopoiesis, and tumor repression (1-4).

Function and Interaction

CHD2 plays an important role in DNA damage response signaling, and mutations in CHD2 give rise to the accumulation of γH2AX (which is a chromatin-associated DNA damage response mediator) following DNA damage and to deficiencies in repairing DNA damage caused by ionizing and ultraviolet radiation, thus indicating that CHD2 is essential for maintaining genomic stability (1,5). Mutations in CHD2 are reported to affect the development and survival of mice by delaying embryogenesis and increasing perinatal lethality (6).

Disease Association

CHD2 has been shown to be involved in tumor suppression, and mutation of CHD2 can result in greater susceptibility to the formation of lymphoid tumors (1). Furthermore, CHD2 is a candidate tumor repressor in classical Hodgkin’s lymphoma (cHL) (7).
