Introduction

Full name: Metal Response Element Binding Transcription Factor 2. MTF2 is a functional component of Polycomb repressive complex-2(PRC2) which mediates transcriptional repression of target genes,including the Hox gene cluster and the Cdkn2a genes,is expressed as four different isoforms and the longest one contains a Tudor domain and two plant homeodomain (PHD) fingers(1-2).MTF2 specifically binds histone H3 trimethylated at Lys-36(H3K36me3) and recruits the PRC2 complex,leading to enhance PRC2 H3K7me3 methylation activity(3),is involved in transcriptional regulation during embryonic stem cell self-renewal and differentiation(4).

Function and Interaction

MTF2 forms a complex with PRC2 and binds to Hox genes in a PRC2-dependent manner,also exhibits a profound synergistic effect on PRC1-mediated Hox gene expression(1).MTF2 interacts with cdk PHO85 in vivo and in vitro and form a kinase complex that has G1-periodic activity,contributes to cell cycle progression(5).MTF2 is required for gene regulatory networks which controls self-renewal and commitment in embryonic stem cells(6-7).
