Introduction

Full name: Chromobox protein homolog 3. Cbx3 is a member of the family of heterochromatin protein 1 (HP1) proteins,which is a family of highly conserved heterochromatin-associated non-histone chromosomal proteins and has been reported to specifically recognize methylated histone H3K9 mark.Cbx3 contains an N-terminal chromodomain (CHD) and a C-terminal dimerization chromo shadow domain (CSD) separated by a poorly conserved hinge region(1-3).

Function and Interaction

Cbx3 is a paralogue of heterochromatin protein 1 (HP1) proteins,posseses two conserved domains,the N-terminal chromodomain (CHD) which selectively recognizes methylated histone H3K9 mark,and the chromoshadow domain (CSD) through which HP1 forms dimers with other HP1 molecules or with other proteins and regulates the binding activity of the CHD(4-5).Cbx3 is involved in silencing at E2F- and MYC-responsive genes in G0 cells(6).Cbx3 is required for the repression of the progestin responsive MMTV promoter in breast cancer cells(7).In conjunction with SUV39H1,Cbx3 is also responsible for chromatin-mediated repression of the HIV-1 gene(8).
