Introduction

Full name: PHD Finger Protein 19. PHF19 contains two isoforms,the long isoform contains an N-terminal TUDOR domain and two PHDs(plant homeodomains) while the short isoform lacks the second PHD finger(PHD2)(1).PHF19 associates with core subunits EZH2,SUZ12 and EED of PRC2,is a component of PRC2(1-5).PHF19 binds trimethylated histone H3 Lys36(H3K36me3) via its TUDOR domain and associates with H3K36me3 demethylase NO66,which is required for recruit the PRC2 complex and NO66 to stem cell genes during differentiation that mediates H3K27me3 methylation,followed by de novo silencing(2-4).

Function and Interaction

PHF19 has a pivotal role in silencing active promoters by directly targeting the PRC2 complex and H3K36 histone demethylases,is required for mES cell differentiation(2-4).PHF19 is a critical regulator of gene repression and mES cell self-renewal that acts by controlling PRC2 binding to target genes,particularly CpG islands regulated by PHF19(6).PHF19 plays an important role in the phenotype switch of melanoma cells from proliferative to invasive states(7).
