Introduction

Full name: Chromobox protein homolog 7 CBX7 is a homolog of Polycomb proteins,contains a chromodomain which is capable of recognizing repressive marks such as trimethylated Lys-9 and Lys-27,respectively,on histone H3(1).CBX7 is a component of Polycomb repressive complex 1 (PRC1),a key regulator of developmental genes.CBX7 chromodomain also binds to trimethylated SETDB1 peptides with equal or greater affinity than histone peptides and don't bind to the non-methylated pepetides,suggests that CBX7 are likely to have an alternative non-histone binding activity(2).

Function and Interaction

CBX7 is the predominant PRC1-associated CBX subunit in proliferating ESCs and requires the H3K27me3 marks to localize to chromatin and thereby silence the expression of lineage commitment genes(3).CBX7 extends the lifespan of a wide range of normal human cells and immortalizes mouse fibroblasts by downregulating expression of the Ink4a/Arf locus and can function independently of BMI-1(4-5).CBX7 can inititate T cell lymphomagenesis and cooperate with c-Myc to produce highly aggressive B cell lymphomas(6).

Disease Association

CBX7 is up-regulated in follicular lymphoma and possesses oncogenic properties in the hematopoietic compartment(6).
